What should Hero do? It'd be really helpful if you provide a thorough explanation/thought process behind the answer.

Is he a calling station?
Does he play most of his hands in position?
Is he normally aggressive?
How has the Hero been playing?
Have you been going to showdown and winning?
Did you pick up any tells from the SB?

You can just bet small for value and protection. The SB misses this board a tonne because they have barely any 6x in their range. This is different to vs BB where they have a big 6x advantage (since their defending range is so wide) and we'd have to be more cautious.

But I also see that Hero doesn't have that much 6's in his range, is this true? All I see is 66 and very slim 65s, which is almost nothing compared to the rest of his wide range. I'm even tempted to think that Villain's range has more 6's just because Hero opens wide and with not many 6's.

CO should have slightly more 6x in the opening range than SB's flatting range, but you're right neither player has much 6x.

You have the stronger range overall though with pockets pairs that SB will lack, AK KQ that SB lacks and also just hands like AT which are doing fairly well vs his flats. So you have overall range advantage and some nut advantage = bet frequently.

Against the BB you would have the overall range advantage but a huge nut disadvantage in terms of 6x, leading to a much more cautious strategy overall

You always have some information. Race, age, dress, body language, facial expression, how attentive he is, if he is doing something other than playing the hand (like drinking alcohol).

I play a live, exploitive, game and it sounds like you want the GTO answer, which I will leave to others.

I think the SB range contains a lot of 6x hands.
Calling the SB usually looks like a weak play, so I would start with a small bet on the flop against an unknown opponent.
The board is very dry and it is difficult for the opponent to defend.
In addition, if the opponent calls us, we will almost always see all the cards on the board, which increases the realization of the equity of our hand.
