PokerStars, Hold'em No Limit - $0.02/$0.05 - 5 players
UTG: $5.23 (105 bb)
CO: $5.00 (100 bb)
BU: $6.91 (138 bb)
SB: $6.73 (135 bb)
BB (Hero): $5.75 (115 bb)
Pre-Flop: ($0.07) Hero is BB with J♣ Q♣
2 players fold, BTN raises to $0.15, 1 fold, Hero calls $0.10
Flop: ($0.32) Q♥ 2♥ 5♦ (2 players)
Hero checks, BTN bets $0.15, Hero calls $0.15
Turn: ($0.62) 7♣ (2 players)
Hero checks, BTN bets $0.29, Hero calls $0.29
River: ($1.20) T♥ (2 players)
Hero bets $0.35, BTN raises to $1.90, BB (Hero) folds
Does a donk bet R of 1/3 pot ever make sense? I wanted to make a block bet here so I wouldn't call for 2/3 and fold on the raise.

I'd be very surprised if there was even a non zero percent of lead OTR, range asymmetry just doesn't allow for it

What conditions must be met for donk beta in block bet form? Is a block bet only used against checkback bets and lead hero bets?

95% of donk bets are ~1/3
Conditions for donk - range asymmetry (usually for the ~75th equity percentile plus aka nut adv)
Common donk bet spots include:
°Low connected boards whereby PFC has a higher density or exclusive straights
°when PFA bets high freq' and we have a higher density of a particular nuttish hand class OTT (usually paired/ FC turns) (higher freq' of leads when we fold unusually more than 1-A although this is somewhat countered by the fact that when we overfold it's due range asymmetry aka PFA has larger than normal nut adv)
°OTF on the Paired boards (usually BB v EP) where we have a significant trips ADV (usually donk freq' reduces as the numerical height of the pair increases)
Note: most simplify for no leads BB v BU as although it is fairly prevalent at equilibrium, we don't really gain much, if any, EV. This is due to range asymmetry not being as dramatic as other formations and holistically its quite easy for PFA to over-cbet a spot we lead high freq' yet have simplified for range check.
We can see a donking spot like we've muddled up IP & OOP's ranges in the solver.
