My opponent declares gin and lays down her cards.

I have three runs and the lone left over card is an Ace, which I can play on her three aces.

Does she still get 25 points for Gin?

You don't get to lay off if the opponent declares a Gin, only after they knock.

There are two variations of Gin Rummy that would allow the Ace to be laid off.

Lay off Gin Rummy and Super Gin Rummy. The details of these variations can be found in Scarne on Card Games.

But if you are playing Standard Gin Rummy then no the Ace cannot be laid off and opponnent receives 25pts for Gin plus the 1 pt in your hand.
