At 50PLO about how much volume would I need? I l ow I may need to mix other stakes as well to keep volume.

Would ~40k hands a week at 50PLO get me to the 1k tier?

How loose are you. 40k hands at 50PLO will likely be enough or close to the 1k bracket. Would be about 20k points.

40k hands in a week sounds absurdly high but then I remembered the time I did like 55k in a day on FTP rush.
