I just sat down only 1 an hour ago. V is unknown and I label him as "LAG". I saw that he semi-bluffed with a flush draw and barrelled the river (in position) when he missed his draw.

I'm a TAG/NIT, but V probably doesn't observe anyway. I have $950 behind and V has me covered.

Mandatory straddle and I open to $55 with QQ V calls in CO and blinds fold.

($130) flop is Q69 I lead out pot and V calls

($390) Turn is Q I bet $130 and V calls

($650) River is a 2

Hero? was my sizing on the turn negative/positive EV?

You have quads. You want to get stacks in. Bet at least 200 on the turn and gii on the river.

This is a pretty wet board. When villain calls a pot sized bet on the flop it's most likely he is drawing to something.

Turn bet should be someplace between $250 and $300. You have a monster hand and want to get all in at some point. Villain can't have any QX so they have fewer call down hands putting more draws in their range. It's likely the turn is the last chance you have to make money against the draws.

As played shove river and hope villain has a boat. Missed draws mostly won't call anything and the boats will call everything. Your small turn bet left you with an awkward pot sized shove but since villain can't have QX don't focus on trying to draw in villains medium hands. It's also a bad situation to try to induce a bluff by checking, villain is likely to realize you likely are trapping with QX.
