is there a calculation behind 3betting a % of hands vs a open %?
example:
button opens 40% and sizing is 3bb => sb 3b x % and sizing 10.5bb

Its a complex situation. Also depends on stack sizes, type of game (NL, FL, PLO, etc)

No. You can only do it by solving poker completely, by which I mean tabulating the EV of every hand against the opening range, given all possible board runouts and all possible action sequences (checks, bets, raises and folds), and then working backwards from the river to pre-flop, altering the 3-bet range and seeing if you get better results with it.

If the game stopped on the flop, then it is solved already for heads up 2 players.

The problem is that all streets are related, as Arty pointed out, and that every possible run out relates back to the very first action preflop.

You can’t chop the game into subgames and solve the entire game by being subgame perfect, but an AI that uses such methods is ridiculously hard for a human to compete against.
