Just avoid off strip poker rooms like the Orleans or Red Rock which can be filled with nitty locals. (or so I've been told)
Some places might give you a discount on your room if you clock in 6-8 hours of play but generally speaking there's not a lot of benefit to playing in one room if you're going to clock in less than 50 hours on your card.
I'd say play in as many rooms as possible if you're new to Vegas just to get the experience and enjoy each atmosphere.
