For someone who’s studied preflop sims of this, what are the main differences in strategy between for instance 6 players with 40bb stacks and a table with 1 40bb stack and 5 100bb stacks?

My guess is the deeper stacks have to be tighter while the short stacker can be looser

And who has the highest range EV across the average of being in all positions?

Again my guess would be the shortest stack but would be very interested to hear from someone who’s seen any sims

I believe Zkesic has data regarding this if he is willing to share it; though, I do believe he made a post with similar questions/answers in his PGC.

Typically you wouldn't have specific ranges for this situation, but you have ranges for 100BB 6 handed and ranges for 40BB 6 handed.

So what you should do is assign to the shortstack player a shorstack range, and to the big stacked players a hand from the range. Unless the only 2 players left are someone with 40BB and someone with 100BB, in which case the effective stack is now 40BB for both players. It would be reasonable to assume that with 3 players, the effective stack could be somewhere in the middle I guess?

Current GTO is about starting with a general base and knowing how to approximate the specific situation to our general data.

The question doesn't quite make sense, what did you mean?
Range and EV are two separate things, not sure what range EV is.
Range is a matrix, a set of hole cards (of which there are 1326 combinations) with a frequency assigned to each. So in itself it wouldn't be something that's orderable from highest to lowest, because it has more than one dimension.
EV is orderable and comparable yes, but no player has an EV here, I guess the player who plays the best has the most EV?

If the question is whether either player holds an advantage? If you don't factor rake, there should be no advantage is my guess. Unless you consider that players start with a forced bet, so logically if the player starts with 1BB, then they have a disadvantage, as other players have the option to fold against a forced all in, but this becomes negligible when reaching 40BB.
